White root rot, caused by the soil-borne ascomycete Dematophora necatrix, is a highly destructive disease affecting perennial crops worldwide. This polyphagous pathogen attacks a vast range of hosts, including pome fruits, stone fruits, grapevines, and olives. The infection is characterized by a conspicuous white, cottony mycelial mat that envelops the roots, leading to rapid cortical decay and vascular collapse. Due to its ability to survive for years on organic debris, D. necatrix causes recurring infections that are exceptionally difficult to eradicate. 

The epidemiology of D. necatrix is driven by high soil moisture, spreading through root-to-root contact or contaminated soil movement. A critical management challenge is that the pathogen often remains asymptomatic in the early stages, with visible symptoms like wilting and dieback appearing only after extensive colonization. Consequently, early molecular detection is vital for orchard biosecurity. Identifying the pathogen’s DNA within soil or asymptomatic tissues enables timely quarantine and localized eradication, preventing widespread loss of high-value plantations.
