Aphelenchoides ritzemabosi is a highly destructive foliar nematode that poses a significant threat to the ornamental plant industry, most notably affecting chrysanthemums and various other nursery crops. As a migratory endoparasitic nematode, it is the primary causal agent of angular leaf spots and systemic decline in over 200 plant species. Due to its environmental resilience and status as a regulated pathogen, A. ritzemabosi is a critical concern for growers, as infestations often lead to complete loss of aesthetic value and significant reductions in marketable yield. The infection is characterized by distinct, vein-bounded necrotic lesions on the leaves, which transition from yellow to dark brown as the tissue dies. These nematodes migrate across plant surfaces within films of water, entering through stomata to feed on mesophyll cells. A key feature of A. ritzemabosi is its ability to enter a dormant state (anhydrobiosis) within dried plant debris, allowing it to persist for several years until favorable conditions return. Because early infestations can be latent or masked by other physiological stresses, rapid and sensitive molecular detection is essential for phytosanitary certification and the maintenance of healthy nursery stocks. This nematode can be detected using Real-Time PCR instruments with this specially designed kit, targeting its genomic DNA. The assay utilizes gene-specific primers and a hydrolysis probe based on TaqMan® technology. Specifically, the primers and probe represent 100% homology with over 95% of the NCBI database reference sequences available at the time of design.  Along with the simple protocol, this feature makes it easy to obtain quick, reliable and rapid results to its users.
