Moniliasis, also known as brown rot, is a major fungal threat to stone and pome fruit production, causing substantial losses from bloom to post-harvest. The Letgen Biotech Multiplex Real Time PCR assay utilizes a targeted molecular approach for the simultaneous detection and differentiation of the two primary causal agents: Monilia laxa and Monilia fructigena.

The epidemiology of these pathogens is driven by high humidity and spring rainfall. Both fungi overwinter in mummified fruits and infected branch cankers. During the growing season, conidia are dispersed by wind, rain, and insects, invading the host through floral parts or fruit injuries. Once established, they cause rapid tissue necrosis, which often progresses into systemic wood infections if not managed early.

Monilia laxa is the principal agent of Blossom and Twig Blight. It primarily attacks flowers during bloom, causing them to wilt and turn brown rapidly. The infection often travels into the twigs, resulting in small cankers and terminal dieback, which reduces the overall fruit-bearing capacity of the tree.

Monilia fructigena is primarily responsible for Fruit Brown Rot. It induces characteristic circular, brown, soft lesions on the fruit surface, often accompanied by concentric rings of yellowish-gray conidial tufts. Infected fruits eventually dehydrate and shrink, becoming “mummies” that serve as a persistent inoculum source for the following season.

Because these pathogens often occur concurrently in the same orchard and produce overlapping symptoms, visual diagnosis is often unreliable for species-specific management. The multiplex assay enables the simultaneous detection of pathogen-specific DNA targets in a single reaction, supporting an efficient and reliable workflow This molecular approach allows for high-precision fungicide timing and sanitation strategies, ensuring protection during the critical bloom and ripening periods before irreversible yield loss occurs.
