Powdery mildew is a highly prevalent fungal disease that severely impacts fruit quality and tree vigor in pome and stone fruit orchards. The Letgen Biotech Multiplex RT-qPCR assay utilizes a targeted molecular approach for the simultaneous detection and differentiation of its two primary agents: Podosphaera leucotricha and Sphaerotheca pannosa.

The epidemiology of these pathogens is centered on their ability to overwinter as dormant mycelia within infected buds and, in some cases, as sexual fruiting bodies (chasmothecia). As obligate biotrophs, they initiate primary infections on emerging tissues as soon as buds break in the spring. Warm temperatures and high humidity trigger the rapid production of wind-dispersed conidia, facilitating explosive secondary spread before symptoms become visually prominent.

Podosphaera leucotricha primarily targets apples and pears, covering young shoots with a white, powdery coating. This infection stunts growth and induces “russeting” on the fruit surface, drastically reducing marketable yield.

Sphaerotheca pannosa is the major causal agent in peaches, nectarines, and roses. Beyond causing leaf chlorosis, it forms white, felt-like patches on the fruit that render the crop unmarketable.

Because these pathogens often co-exist in mixed orchards and produce nearly identical white-powder symptoms during early stages, visual diagnosis is often insufficient for precise management. The Multiplex assay enables the identification of pathogen-specific DNA in a single reaction, streamlining the diagnostic workflow. This molecular approach allows for optimal fungicide timing and resistance management, ensuring protection during the critical early-season growth stages before systemic infection compromises orchard productivity.
